RESUMEN

Acanthocephalosis is caused by the endoparasite Neoechynorhyncus buttnerae and affects fish farmed in the Amazon. This study assessed the efficacy of therapeutic levamisole hydrochloride (LVC) baths against N. buttnerae and its effects on juvenile tambaqui blood parameters. In vitro and in vivo tests were carried out, the latter employing two experimental therapeutic LVC bath protocols. Concerning in vitro efficacy, the T75 (75 mg.L-1 LVC) and T100 (mg.L-1 LVC) treatments were 100% effective in 15 min, while the T50 (50 mg.L-1 LVC) and T25 (25 mg.L-1 LVC) treatments required parasite exposure for 45 and 60 min, respectively. During exposure, the parasites displayed reduced motility, proboscis retraction, coiling into a spiral shape, body rigidity and swelling. The LVC LC50-72h for juvenile tambaqui was 115 mg.L-1 . Regarding in vivo efficacy for Protocol I (8-h bath), the T125 resulted in 82% effectiveness, while in Protocol II (two 8-h baths with a 24-h interval), the T115 treatment (115 mg.L-1 LVC) achieved 95.6% effectiveness without clinical intoxication signs, despite behavioural changes. No significant changes were observed in fish blood parameters. LVC was, therefore, highly effective both in vitro and in vivo in controlling the acanthocephalan N. buttnerae without compromising tambaqui juvenile homeostasis.

Myxosporea parasitize many organs in fresh and saltwater fish. Species of the genus Myxobolus parasitizing the gills and other organs of the tambaqui Colossoma macropomum have been described. In the present study, blood smears were made from juvenile tambaqui and were stained with May Grunwald-Giemsa-Wright in order to identify myxozoan parasites. Out of a total of 36 fish examined, one specimen (2.7%) that was reared in a cage presented spores that were identified as M. colossomatis, whereas fish kept in 250 L tanks showed prevalence of 5.5%. This is the first report of M. colossomatis in the blood of farmed tambaqui in the Amazon region. These results indicate that myxozoan parasites should also be investigated in fish blood smears. Some myxosporean species may cause diseases in fish, and these species need to be identified so that adequate preventive sanitary control can be instituted.

A criação de peixes carnívoros nativos vem recebendo grande atenção devido ao promissor uso na aquicultura. Entre estes peixes destacam-se os tucunarés do gênero Cichla, de grande interesse para a pesca esportiva e a indústria de peixes ornamentais da Amazônia. O presente estudo descreve a relação peso-comprimento, fator de condição relativo (Kn) e parâmetros sanguíneos do tucunaré Cichla temensis Humboldt, 1821 cultivado na Amazônia central. Os peixes submetidos a treinamento alimentar na alevinagem receberam ração extrusada contendo 45% de proteína bruta na engorda e tiveram Kn variando de 0,925-1,199, indicando boas condições de higidez no cultivo. A equação obtida da relação peso-comprimento foi P = 0,0073.Ct3,1435, indicando crescimento do tipo isométrico, que é o mais desejável para peixes em uma piscicultura. O número de eritrócitos, trombócitos e leucócitos totais, hematócrito, concentração de hemoglobina, volume corpuscular médio (VCM), concentração da hemoglobina corpuscular média (CHCM), linfócitos, monócitos e neutrófilos mostrou variação intra-específica. Foi observada correlação positiva significativa (p<0,001) do número de eritrócitos totais com a concentração de hemoglobina e o hematócrito. Estes foram os primeiros resultados sobre parâmetros sanguíneos de C. temensis e poderão ser usados como referência para comparação em estudos futuros para avaliar o estado de saúde deste peixe em diferentes ambientes, pois a medida destes parâmetros pode ser usada como ferramenta rápida para diagnóstico de doenças, estresse ou desnutrição.


Farming of native carnivore fish species has drawn attention due to their promising use in aquaculture. Among these species, tucunaré of the genus Cichla stand out, them being of high economical interest for sport fishing and Amazon’s industry of ornamental fish. The present study describes the weight-length relationship (WLR), relative condition factor (Kn), red blood cell parameters, thrombocytes and leukocytes count of Cichla temensis Humboldt, 1821, farmed in central Amazon. Fish that underwent food training during fingerling culture received extruded ration containing 45% of crude protein during fattening, and had Kn with values from 0.925-1.199, which indicated good health condition during the culture. The equation obtained from the WLR was W = 0.0073Lt3.1435, indicating an isometric growth, which is the desirable for fish of fish farm. Red blood cell counts, total thrombocyte and leukocyte counts, hematocrit, hemoglobin concentration, mean corpuscular volume (MCV), concentration of mean corpuscular hemoglobin (MCHC), lymphocytes, monocytes and neutrophils had intra-specific variation. A significant (p<0.001) positive correlation of the red blood cells number with the hemoglobin concentration and hematocrit was found. These are the first sets of blood parameters for C. temensis and could be used as reference for comparison in further studies to evaluate the health status of this fish in different environments, because assessment of these parameters may be used as quick tool for diagnosing diseases, stress and malnutrition.

Mixosporídeos parasitam diversos órgãos de peixes de água doce e salgada, tendo sido descrita espécie do gênero Myxobolus parasitando brânquias e outros órgãos de tambaqui Colossoma macropomum. No presente trabalho, extensões sanguíneas de juvenis de tambaquis foram confeccionadas e coradas com May Grunwald-Giemsa-Wright para identificação de parasitos myxozoários. Do total de 36 peixes examinados, um espécime (2,7 por cento) cultivado em tanque-rede apresentou esporos identificados como M. colossomatis, enquanto os peixes mantidos em tanques com 250 L apresentaram prevalência de 5,5 por cento. Esse é o primeiro registro de M. colossomatis no sangue de tambaqui cultivado na Amazônia. Esses resultados indicam que parasitos Myxozoa devem ser também investigados em extensões sanguíneas. Algumas espécies de mixosporídeos podem causar doença em peixes, sendo necessária a identificação da espécie para um adequado manejo sanitário preventivo.

In order to determine the lethal dose (96-h LD50) of the bacteria Aeromonas hydrophila to matrinxã, Brycon amazonicus, to be applied in challenge tests, 90 fish (63.23 ± 6.39 g) were divided into five treatments, with different bacterial solutions: T1 - Control (0.9 percent NaCl saline solution); T2 (4 x 10(11) cells/ mL); T3 (5 x 10(11) cells/ mL); T4 (1.36 x 10(12) cells/ mL) and T5 (3.06 x 10(12) cells/ mL). Fish were previously anesthetized with benzocaine (60 mg L-1), inoculated in the peritoneal cavity with the bacterial suspensions and then distributed into fifteen 80-L test chambers, where the water variables were monitored and fish mortality was observed. The experiment was randomly designed in three replicates and the 96-h LD50 was estimated according to the trimmed Spearman-Karber method. Water quality variables remained within adequate ranges for fish health and performance. Fish mortality rate increased with the bacterial concentrations of A. hydrophila (T1 = 0 percent; T2 = 16.66 percent; T3 = 44.44 percent; T4 = 72.22 percent and T5 = 100 percent), and the first mortalities were observed after 57 h, although the signs of the bacterial infection were already observed 24 h after the inoculation. The results indicate that the 96-h LD50 value of A. hydrophila to matrinxã is 6.66 x 10(11) cells/ mL.


Para determinar a dose letal (DL50 96-h) da bactéria Aeromonas hydrophila para o matrinxã, Brycon amazonicus, com aplicabilidade para testes de desafio, foram utilizados 90 peixes (63,23 ± 6,39 g), divididos em cinco tratamentos, com diferentes soluções bacterianas: T1 - Controle (solução salina 0,9 por cento NaCl); T2 (4 x 10(11) células/ mL); T3 (5 x 10(11) células/ mL-1); T4 (1,36 x 10(12) células/mL-1) e T5 (3,06 x 10(12) células/ mL-1). Os peixes foram previamente anestesiados com benzocaína (60 mg L-1), inoculados na cavidade peritoneal com as suspensões bacterianas e distribuídos em 15 aquários de vidro de 80 L de capacidade, com aeração constante. O experimento teve duração de 96 h, no qual foram monitoradas a mortalidade e a qualidade da água. O delineamento experimental foi inteiramente casualisado com três réplicas e a DL50 96-h foi estimada de acordo com o método Spearman-Karber. Durante o experimento os parâmetros físico-químicos da água permaneceram dentro das condições consideradas adequadas para o desenvolvimento e saúde dos organismos aquáticos. A mortalidade dos peixes aumentou nas concentrações crescentes de A. hydrophila (T1 = 0 por cento; T2 = 16,66 por cento; T3 = 44,44 por cento; T4 = 72,22 por cento e T5 = 100 por cento), contudo, as primeiras mortalidades ocorreram em 57 h após a inoculação das concentrações bacterianas, sendo observados os primeiros sinais de infecção em 24 h após a inoculação. Os resultados indicam que o valor da DL50 96-h da bactéria A. hydrophila para o matrinxã foi igual a 6,66 x 10(11) células/mL de solução salina.

The present study evaluated the effectiveness of Amazonian nut (Bertholletia excelsa) as an alternative source of vegetal protein in tambaqui (Colossoma macropomum) diet. Performance and physiological status of fish fed for 60 days were evaluated. Four experimental isonitrogenous diets with 36 percent crude protein were formulated with increasing levels of nut meal (0, 10, 20 and 30 percent). Results showed the same growth performance for fish fed with diet with different levels of Amazonian nut than that without this ingredient (control). Analysis of physiological parameters (hematocrit, erythrocyte number, hemoglobin concentration, hematimetric indexes, total plasma protein and plasma glucose) corroborate these results, with no significant differences among treatments. Therefore, adding up to 30 percent of Amazonian nut in tambaqui diet there is no negative effect on physiological homeostasis and growth performance, indicating that the Amazonian nut is a promising alternative dietary protein source ingredient for tambaqui.


Este estudo avaliou a eficácia da farinha de castanha da Amazônia (Bertholletia excelsa) como fonte alternativa de proteína vegetal na dieta do tambaqui (Colossoma macropomum). Para isso, foram avaliados o desempenho e o estado fisiológico dos peixes alimentados durante 60 dias. Foram formuladas quatro dietas experimentais isoproteicas com 36 por cento proteína bruta, com níveis crescentes de farinha de castanha (0, 10, 20 e 30 por cento). Os resultados mostraram que os diferentes níveis de castanha da Amazônia mantiveram o mesmo desempenho zootécnico obtido para os peixes alimentados com dieta sem esse ingrediente (controle). Esses resultados são corroborados pela análise dos parâmetros fisiológicos: hematócrito, número de eritrócitos, concentração de hemoglobina, índices hematimétricos, proteínas plasmáticas totais e glicose plasmática, os quais não demonstraram diferenças significativas relacionadas aos diferentes tratamentos. Portanto, até 30 por cento de farinha de castanha da Amazônia pode ser adicionada à ração sem comprometer a homeostase fisiológica e o desempenho do tambaqui, mostrando-se como um promissor ingrediente alternativo de fonte proteica na dieta para essa espécie.

O presente estudo comparou os níveis plasmáticos de glicose, proteínas totais, uréia, triglicerídeos e colesterol, a contagem de eritrócitos, leucócitos e trombócitos entre alevinos e juvenis de Arapaima gigas (Osteoglociformes, Arapaimidae) de uma piscicultura semi-intensiva de Manaus, estado do Amazonas, Brasil. Os alevinos de A. gigas apresentaram níveis significativamente (p<0,05) menores de proteínas totais, triglicerídeos, uréia e Volume Corpuscular Médio (VCM) e maior concentração de glicose, colesterol e hemoglobina, contagem de eritrócitos, hematócrito e Concentração da Hemoglobina Corpuscular Média (CHCM) quando comparados aos juvenis. Porém, não houve diferença significativa (p>0,05) no número de trombócitos e leucócitos totais entre alevinos e juvenis. Os juvenis de A. gigas apresentaram menor quantidade de linfócitos e maior quantidade de monócitos, neutrófilos e eosinófilos, quando comparados aos alevinos. Em estudos futuros, espécimes de A. gigas com outras idades também deverão ser comparados para melhor entendimento dos efeitos da idade neste peixe amazônico, pois estas informações, além de possibilitarem uma melhor compreensão da fisiologia desta espécie, poderão auxiliar no estabelecimento de estratégias para seu manejo.


The present study compared the biochemical and hematological parameters of fingerlings and juveniles Arapaima gigas (Osteoglociformes, Arapaimidae) from a semi-intensive fish farming in Manaus, Amazon State, Brazil. Fingerlings A. gigas had lower levels of total protein, triglycerides, urea and mean cell volume (MCV), but higher (p<0.05) glucose, cholesterol and hemoglobin concentration, red blood cell counts, hematocrit and cell hemoglobin concentration (MCHC) when compared to juveniles. However, there was not significant difference (p>0.05) in the total thrombocytes and total white blood cell counts between fingerlings and juveniles. Juveniles A. gigas presented lower lymphocytes number and higher monocytes, neutrophils and eosinophils number when compared to the fingerlings. In further studies, A. gigas specimens with other ages should also be compared for a better understanding of the ontogenic development in this Amazon fish, because this information besides increase the knowledge of the physiology of this species, can aid in the establishment of strategies for its adequate handling.

The aim of this study was not only to determine the red blood cells parameters, thrombocyte and leukocyte counts in farmed Brycon amazonicus (matrinxã), to compare these parameters among Bryconinae species from literature, and also to investigate the presence of special granulocytic cells in these fish. The results of the blood cells parameters here established for farmed B. amazonicus, a species of great economic importance in Brazilian aquaculture, could help a better understanding of the blood features in natural populations of this Amazon species. Blood parameters varied between Bryconinae species investigated, mainly the red blood cell counts, hemoglobin, hematocrit and mean corpuscular volume (MCV). The presence of the blood granulocytes, neutrophils and heterophils in matrinxã suggest that both leukocytes can be a characteristic for Bryconinae family. Furthermore, it indicates that the existence of special granulocytic cells in the blood of Bryconinae species from literature is an artifact, and this was herein discussed.


O objetivo deste estudo foi determinar os parâmetros eritrocíticos, as contagens de trombócitos e de leucócitos de espécimes de Brycon amazonicus (matrinxã), criados em cativeiro e compará-los com aqueles descritos na literatura para outras espécies de Bryconinae. Além disso, foi ainda investigada a presença de células granulocíticas especiais nestes peixes. Os resultados dos parâmetros sangüíneos apresentados para B. amazonicus podem ajudar a entender melhor as características sangüíneas em população natural desta espécie de grande importância para a aqüicultura brasileira. Os parâmetros sangüíneos das espécies de Bryconinae investigadas apresentaram variação interespecíficas principalmente a contagem de eritrócitos, hemoglobina, hematócrito e volume corpuscular médio (VCM). A presença dos granulócitos sangüíneos, neutrófilos e heterófilos em matrinxã sugere que esta pode ser uma característica das espécies pertencentes à subfamília Bryconinae. Além disso, indica que a existência de células granulocíticas especiais no sangue de espécies de Bryconinae da literatura é um artefato, e isto foi aqui discutido.

Poor water quality condition has been pointed out as one of the major causes for the high mortality of ornamental fishes exported from the state of Amazonas, Brazil. The purpose of the current study was to define water quality standards for cardinal tetra (Paracheirodon axelrodi), by establishing the lower and higher for lethal temperature (LT50), lethal concentration (LC50) for total ammonia and nitrite and LC50 for acid and alkaline pH. According to the findings, cardinal tetra is rather tolerant to high temperature (33.3 ºC), to a wide pH range (acid pH=2.9 and alkaline pH=8.8) and to high total ammonia concentration (23.7 mg/L). However, temperatures below 19.6 ºC and nitrite concentrations above 1.1 mg/L NO2- may compromise fish survival especially during long shipment abroad.


A má qualidade da água tem sido apontada como uma das maiores causas da alta mortalidade dos peixes ornamentais exportados pelo Estado do Amazonas, Brasil. A proposta deste estudo foi definir padrões de qualidade da água para o cardinal tetra (Paracheirodon axelrodi), estabelecendo a menor e a maior temperatura letal (LT50), a concentração letal (LC50) para amônia total e nitrito e LC50 para pH ácido e alcalino. De acordo com os resultados, o cardinal tetra é mais tolerante a temperaturas elevadas (33,3 ºC), a amplos limites de pH (pH ácido = 2,9 e pH alcalino = 8,8) e a alta concentração de amônia (23,7 mg/L). Entretanto, temperaturas abaixo de 19,6 ºC e concentrações de nitrito acima de 1,1 mg/L NO2- podem comprometer a sobrevivência dos peixes, especialmente durante longos períodos de transporte para o exterior.

Juvenile matrinxã (Brycon amazonicus) were fed with different dietary vitamin C (ascorbic acid or AA) concentrations for 2 months in order to evaluate its effect on the physiological condition of this species. Five treatments were tested: Reference diet-RD (350 mg AA kg(-1) and 45% crude protein commercial feed); C400, C500, C600 and C800 with supplementation of 400, 500, 600 and 800 mg AA kg(-1). Fish fed 800 mg AA kg(-1) supplemented diets presented greater weight gain and survival. Hematocrit, hemoglobin concentrations and erythrocytes showed significant increase (p<0.05) in C600 and C800 relatively to the others. Total leukocyte values were significantly higher in fish fed vitamin C supplemented diets relatively to those fed RD and leukocytes count of C600 and C800 were the highest among the treatments. Monocytes were the only cells of fish defense mechanism that presented variation, and C600 and C800 presented the lowest values (P<0.05). Glucose level was significantly different relatively to RD only in C400 and the plasmatic total protein values for fish fed vitamin C supplemented diets were higher than for fish fed the RD. Fish plasmatic ions (Na(+) and Cl(-)) were significantly lower for C500, C600 and C800 relatively to C400 and RD. The physiological profile and the growth performance of matrinxã suggest that diet supplemented with 800 mg AA kg(-1) can improve its performance and increase the survival rate.

This study evaluated the influence of diets supplemented with 500, 800, 1200 mg kg-1 of vitamin C (ascorbic acid or AA) and vitamin E (alpha-tocopherol or alpha-T) on the physiological responses of pirarucu fed for 2 months. Weight and mortality were not affected by dietary vitamin type or their concentrations. Significant increase (p<0.05) on the red blood cells count was obtained on treatments with 800 and 1200 mg AA kg-1 and on the hemoglobin concentration on treatment with 500 mg alpha-T kg-1 relatively to control. Mean corpuscular volume presented a significant decrease (p<0.05) on treatment with 800 and 1200 mg AA kg-1 when compared to control. Mean corpuscular hemoglobin concentration was significantly high (p<0.05) on treatment with 500 mg alpha-T kg-1. Only in vitamin C treatments, we noticed a significant increase (p<0.05) in the number of leucocytes relative to control. All fish in the vitamin-supplemented treatments, except 500 mg AA kg-1, had high total protein values compared to control. Fish treated with 800 or 1200 mg alpha-T kg-1 also showed increases in plasma glucose concentrations. Our results suggest that 800 and 1200 mg AA kg-1 are probably the most suitable concentrations for pirarucu diets, although high vitamin E diets are not necessary for quantitative leucocyte increases for this species.

This study evaluated the efficacy of dietary vitamin C (ascorbic acid or AA), vitamin E (alpha-tocopherol or alpha-T), and C+E supplementation on the blood parameters of Arapaima gigas grown in net cages for 45 days. Four treatments were tested: control (commercial feed); C800; E500 and C+E (800+500) with supplementation of 800 mg AA kg(-1), 500 mg alpha-T kg(-1) and 800+500 mg AA+alpha-T kg(-1), respectively. Hematocrit (Ht), red blood cells (RBC), and hemoglobin concentration (Hb) (oxidative status indicators), thrombocytes and leukocytes (immunological indicators), plasma protein and glucose were evaluated. Fish fed vitamin C and C+E supplemented diets showed greater weight gain and survival. Dietary vitamin C and C+E diet supplementation resulted in increased Ht, Hb, RBC, MCHC, total leukocytes, total proteins, thrombocytes and eosinophils compared to the control and alpha-T. The alpha-tocopherol-supplemented diet reduced the number of total thrombocytes, lymphocytes and neutrophils and increased glucose and eosinophils relatively to the control. In general, leukocytes and thrombocytes were good indicators of the efficiency of vitamin on the defense mechanism of the A. gigas reared in cages. Results indicate that high alpha-T diet supplementation provides no benefit for the maintenance of the oxidative or the immunological status of A. gigas. However, it was demonstrated that high dietary AA improves A. gigas immunological status. Red blood cell indices and immune system indicators showed no synergistic effect between the vitamins after supplementing the A. gigas diet with alpha-T+AA.
